A woman in her 60s was in a critical condition when a bus crashed on Burley Griffin Way in Harden on Friday.
The bus had 30 people on board, including children, and a further 26 patients were being treated for minor injuries.
On Thursday, a 40-year-old woman and two children were being treated by paramedics for minor injuries and shock after their car rolled on the Hume Highway at Bookham.
Those crashes followed three fatalities in separate incidents on roads in NSW last weekend.
Traffic volumes have already increased on major highways in the Yass Valley with the start of the school holidays, Inspector Matthew Hinton of Yass Police Station said.
"Drivers need to heed the warnings; if you feeling tired take a break," he said.
"You might be a little late on your journey but it's better than being in hospital for the next month recovering from serious injuries, or worse still injuring a loved one.
"Fatigue is increasing as a causal factor in accidents in the Yass Valley and these major accidents are occurring on fine sunny days, and with the two recent accidents on the Hume Hwy, mid-morning, after people have been on the road for a couple of hours."
Police will be present on local roads across the long weekend (October 4-7) when double demerits will be in place.
"It's not just your life in your hands but your passengers and other motorists," Inspector Hinton said.
